What is the easiest way to take say take a 30 second sample from a 4 minute
mp3 file and make it a separate mp3 file?

    Audacity: http://audacity.sf.net/

If quality is an issue you might want to find something that doesn't
need to reencode it. But i'm assuming its not.

What an excellent couple of links, and a bonus - the first also leads to
http://mpesch3.de1.cc/#1by1 which is a player that understands cue files.

foobar2000 does this well, it also understands rar/zip/etc archives and
can play any combination of these, eg. a cue+cd image in a rar file, or
a flac file containing a whole CD along with a cue file. The diskwriter
makes this incredibly useful, as it allows you to extract the individual
tracks, even encoding on the fly, with any encoder, if you want.
